I'M A DENTIST AND IN ORDER TO ACHIEVE STERILIZATION YOU NEED 20 PSI AND ABOUT 120-130 C FOR ABOUT 20 MINS IN A AUTOCLAVE.

DR. CAPSLOCK is essentially correct, except for the fact that you NEVER PUT PLASTIC IN AN AUTOCLAVE. For some strange reason it doesn't work out well.

NOT TRUE, SOME PLASTIC IS AUTOCLAVE SAFE.

But you have to check the manufacturer's specs, because if you let your PI and/or lab manager buy things, mistakenly assuming they know what they're doing, they will end up ordering the non-autoclavable stuff, and guess who will be stuck having to clean it up?

(ALSO, I SUSPECT DR. CAPSLOCK IS NOT A REAL DOCTOR. *SHIFTY EYES*)

I hate it when you share labs with morons. (HEY DID YOU BOTHER TO CHECK IF THERE WAS ANYTHING IN THE INCUBATOR BEFORE TURNING THE TEMPERATURE UP? NO? THANKS EVER SO FOR KILLING MY EXPERIMENT, I DIDN'T NEED IT, REALLY.)

I have this problem with someone turning up the speed on the shaking incubator to the highest setting constantly, when the highest setting is way too high for normal bacterial incubation. I think they honestly don't know how to read the RPM dial and just think 'really fast is best, right?' You'd be [not] surprised by how many people set up centrifuges and gas tanks on that principal as well.

They don't even read the "DO NOT CHANGE" notes I stick on the control knob.

Depends on the plastic. I've autoclaved tons of plastic pipette tips and eppendorf tubes, but they're specifically designed to take the heat.

It's not so fun when someone autoclaves something that wasn't, though. Stink-er-roo.
